通過原子轉(zhuǎn)移自由基聚合(ATRP)制備了聚(丙烯酸叔丁酯-b-甲基丙烯酸二甲胺基乙酯)(PtBA-b-PDMAEMA)兩親性嵌段共聚物,在共溶劑THF溶液中加入選擇性溶劑水自組裝制備了PtBA-b-PDMAEMA膠束。以1,2-二(2-碘乙氧烷)乙烷(BIEE)為交聯(lián)劑與PDMAEMA發(fā)生交聯(lián)反應(yīng),制備了殼交聯(lián)膠束(SCL膠束)。通過核磁共振(1H-NMR)、動態(tài)光散射(DLS)和透射電鏡(TEM)研究了殼交聯(lián)膠束的結(jié)構(gòu)和形態(tài),并詳細(xì)探討了BIEE和PDMAEMA的摩爾比對殼交聯(lián)結(jié)構(gòu)的影響。發(fā)現(xiàn)當(dāng)[BIEE]/[PDMAEMA]小于0.5時,易發(fā)生膠束內(nèi)交聯(lián),形成球形膠束;當(dāng)其大于0.5時,傾向于形成膠束之間的交聯(lián),導(dǎo)致其流體力學(xué)直徑的增加。另外,還研究了殼交聯(lián)膠束的pH和溫度響應(yīng)性,發(fā)現(xiàn)隨著溫度和pH的變化,SCL膠束表現(xiàn)出可逆的收縮-伸展性,可以作為藥物控制釋放的良好載體。

The amphipathic block copolymer poly(t-butyl acrylate)-b-poly(2-dimethylamino)ethyl methacrylate (PtBA-b-PDMAEMA) was synthesized via sequential atom transfer radical polymerization (ATRP), and PtBA-b-PDMAEMA micelles were prepared by adding the selective-solvent water into the cosolvent THF solution. The shell cross-linking micelles (SCL micelle) were obtained by cross-linking PDMAEMA using 1, 2-bis-(2-iodoethoxy) ethane (BIEE) as a cross-linker. The structure and morphology of the SCL micelle were investigated by means of 1H NMR, dynamic light scattering (DLS) and transmission electron microscopy (TEM). The effect of molar ratio of BIEE to PDMAEMA ([BIEE]/[PDMAEMA]) on the cross-linking network structure was investigated in detail. It was found that intra-micelle cross-linking tended to be formed when [BIEE]/[PDMAEMA] ratio was lower than 0.5, and the spherical SCL micelles were obtained. When the ratio was higher than 0.5, inter-micelle cross-linking structure would be obtained, which increased the hydrodynamic diameters of the SCL micelles. In addition, the pH/ thermo-responsive behaviors of the SCL micelles were studied. The SCL micelles showed reversible collapse and stretch with the changes of temperature and pH of the medium, and could be good carriers for drug controlled release system.
